Can "water and Spirit" in John 3:5 ever refer to amniotic fluid?

I have been discussing the sacraments etc. with non-Catholics. These people claim that

  1. “Baptism” in the New Testament does not necessarily mean “water baptism”

  2. “water and Spirit” in John 3:5 refer to amniotic fluid then a separate action by the Spirit.

Is there a “definitive” answer I can give them?

I recommend the following:

Immersion Only?

Born Again in Baptism
